Microsoft has identified a problem with Windows 10 Fall Creators Update and certain web apps where they won’t work and will produce a HTTP Error 503.
The cause is a simple one, but overlooked by those testing Fall Creators Update before it was approved. Essentially, the Windows Activation Service is looking for a folder full of files that do not exist. The details are the cause are told in KB4050891.
The fix is to stop the Windows Activation Service and remove the folder completely before starting it up again. Microsoft supplies these steps:

rmdir /s /q C:\inetpub\temp\appPools
net start W3SVC
